Development of a novel membrane-protein specific mammalian display system

At OXGENE we have developed a novel antibody discovery method which involves coexpressing a library of antibodies or antibody mimetics in a population of mammalian cells3 , wherein each cell in the population also displays the target polypeptide on the outer surface. This allows more intractable membrane proteins to be targeted, with representative post translational modifications and displaying epitopes that would be present in a physiological setting. In addition library sizes of up to 109 variants can be routinely screened. This whole process is carried out in CHO cells, in order to reduce risk associated with developability of identified antibodies as biotherapeutics.
